What is method validation?
Every day, people trust that the food they eat, the water they drink, and the medicines they take are safe. This trust, however, relies on a complex system of testing and analysis, in which method validation plays a central role. Method validation is a series of studies that confirm a scientific procedure works correctly for its intended purpose. It also proves that laboratories produce accurate, reliable, consistent results that fit the method’s intended purpose.
We can also refer to this scientific procedure as a method. In fields like analytical chemistry, the method is used to measure the concentration or presence of a specific substance (analyte) in a sample. For example, a laboratory might measure the amount of lead in drinking water, or the level of an active ingredient in a table.
Consequently, method validation is crucial to people’s health and safety. Without it, analytical data can appear misleading, which could potentially cause unsafe products or incorrect regulatory decisions.
Method validation answers questions like:
- Are the actual results close to what the values that they truly should be? (Accuracy)
- If we repeat the test multiple times, will we get the same results each time? (Precision)
- Does the method measure the right substance? (Specificity)
- Can the method detect small amounts of the analyte? (Limit of detection)
As we’ll learn next, the words in parentheses are important dimensions of the method. Let’s examine how method validation covers all of these questions (and more!) in detail.
Method Validation Parameters
A method isn’t just a sequence of steps in an experimental procedure. The procedure is designed to assess several performance characteristics, called parameters. Parameters provide a framework for confirming that a method is reliable and serves its purpose.
In this section, we will cover several key parameters that are commonly evaluated during method validation. These include:
- Accuracy
- Precision
- Specificity
- Linearity
- Limit of detection
- Limit of quantitation
- Robustness
In order for a method to be valid (reliable, consistent, and accurate), it must perform well across all of these parameters. This is what the method validation process sets out to evaluate. But how does it evaluate each of these components individually, and how do they work together to show that a method is valid? Let’s begin our discussion with the parameter of accuracy.
Accuracy
When scientists perform an experiment, it’s important that the results they get are true. Accuracy is how close the obtained test result is to the “true” or accepted value. It tells us whether the method yields the right answer, not just an answer that’s consistent across multiple trials.
Accuracy = Closeness to the true value
Example: The true lead concentration in a sample is 10 mg/L.

- In the table above, note that the obtained results are very close to the true value of 10 mg/L. Since they are close to the true value, the method is accurate.
- However, an obtained result like 8.5 mg/L or 11.5 mg/L would be farther from the true value. In that case, the method would be inaccurate, even if you obtained the same wrong value during every trial.
How do labs evaluate accuracy?
There are a few different approaches that scientists can use to determine how accurate the method is. Here are a few:
- Using certified reference materials (CRMs):
A CRM is a material that has a known (“true”) composition. Because it has a known amount of the same analyte that the experiment is designed to measure, it serves as a standard. Scientists can compare their test results to the CRM’s result. This tells them how close their obtained test results are to the true value. - Performing a recovery test (spike test):
Scientists can add a known amount of the analyte to a sample. (This is called spiking a sample.) Then, they can perform the method procedure to measure the sample’s analyte again. If the procedure recovers a measurement that falls within a certain acceptance range, then the method is considered accurate. Different methods may have different acceptance ranges.- Example: You add 5 mg/L of lead to a sample, then measure again. You obtain a measured value of 4.9 mg/L. Your method’s acceptance range is 95% – 105%, so your measured value must fall within this range of the known analyte spike.
- 4.9 mg/L is 98% of 5 mg/L. You have recovered 98% of the spike. Because 98% falls within the acceptance range of 95% – 105%, your method is accurate.
- Example: You add 5 mg/L of lead to a sample, then measure again. You obtain a measured value of 4.9 mg/L. Your method’s acceptance range is 95% – 105%, so your measured value must fall within this range of the known analyte spike.
Why Accuracy Matters for Consumers
Accuracy ensures that the measured amount of a substance truly reflects the product’s content. In doing so, it protects consumers from food, medicine, and other products that may have been produced using misleading data. It also confirms that medicines provide the correct dose, and that food or water meets safety standards. For example, when a laboratory measures the iron level in drinking water, accuracy ensures the reported value reflects the real concentration. If the test shows a lower value, consumers might unknowingly drink unsafe water.
Using accurate analytical methods in laboratories makes it possible for consumers to trust the quality, safety, and value of the products they use daily. Since accurate methods provide truthful and reliable results, they help consumers make safe decisions.
Precision
It’s important for a method to be consistent and perform the same way each time. This way, scientists know that, even after performing the experiment multiple times, the results obtained from individual trials are trustworthy.
Precision shows how consistent results are when scientists repeat a measurement under the same conditions. Ideally, repeating the test should produce similar values each time. Consistent results give consumers confidence in the information they rely on.
Precision = Repeatability
Precision does not evaluate whether or not the obtained results are correct (close to the true value). That’s what accuracy is for. Instead, precision only evaluates if the obtained results are close to each other. It’s possible for precise values to be either accurate or inaccurate.
Example: The true concentration value of an analyte in a sample is 10 mg/L. You measure the sample five times.
Scenario 1: High Precision

- All of the obtained results are very close to each other, so this method has high precision.
- These obtained results are also close to the true value (10 mg/L), so this method also has high accuracy. Even if the obtained results were very different from the true value, the fact that they are similar to each other across the five trials still shows consistency.
Scenario 2: Low Precision

- Across the five trials, the value of each obtained result varies a lot. They jump all over the place, ranging from 7.5 mg/L to 13.0 mg/L. Because they differ from each other, this method has low precision.
- When the method isn’t very precise, it becomes harder to judge whether or not the method is accurate.
How do labs evaluate precision?
Now, we understand that it’s crucial for a method to be repeatable and consistent. How do scientists determine a method’s precision?
- Repeatability (same scientist, same day):
The same scientist measures the same sample multiple times, each time under identical experimental conditions. If the scientist obtains consistent measurements each time, then the method is considered precise. - Intermediate precision (different experimental conditions):
The same sample is measured multiple times, but each time, a specific variable is changed during the experiment. This might mean that a different scientist measures the sample each time, the experiment is repeated across multiple different days, or a different instrument is used to measure the sample each time. If the obtained measurements are consistent despite the changes, then this indicates that the method is precise. - Reproducibility (different labs):
Two or more scientists in two or more different laboratories perform the same method in the same way as each other. The goal of this is to confirm that the method is reliable in the long term. All of the results obtained by all of the scientists in every lab should be similar to each other.
Why Precision Matters for Consumers
Precision ensures that repeated measurements of the same sample yield very similar results. For consumers, this is reassurance that every batch of medication, food, or cosmetics meets the same specifications as each other. Consumers expect product quality to be consistent every time they buy the product, and precision makes this consistency possible.
For example, when a consumer buys medication, they expect the medication to have the same dose of the active ingredient each time they buy it. If the medication was produced using an imprecise method, then different batches of the medication might have inconsistent potencies, which poses a major safety concern.
- Example: A vitamin supplement claims that each tablet has 100 mg of vitamin C.
- If every tablet contains close to 100 mg of vitamin C, then the consumer will get exactly what they expect. The vitamin supplement was produced using a highly precise method.
- If some tablets have 100 mg of vitamin C, while other tablets have 50 mg or 150 mg, then the supplement’s claim of 100 mg is misleading and potentially unsafe. In this case, the supplement was produced using a method that has low precision.
If a method lacks precision, the measurements obtained will fluctuate unpredictably. For instances, when testing food, a method with poor precision might yield inaccurate measurements of allergen levels in the food. These measurements could put sensitive consumers at risk. Therefore, precise and consistent results protect consumers from health hazards and support public confidence in the product.
Finally, consumers rely on labels and product claims. High precision during testing ensures that the package’s brand and claims accurately reflect the product, which builds trust in the product. Conversely, low precision can lead to recalls, consumer complaints, or long-term damage to a brand’s reputation.
Precision vs. Accuracy
When scientists evaluate a method, they often assess precision and accuracy hand-in-hand. However, they are not identical to each other. A method may be either accurate and inaccurate, and either precise or imprecise.
In the image below, the blue dots represent individual measurements from multiple trials of an experiment. Note the differences between each scenario, and remember that a method should ideally be both accurate and precise.

Specificity
When scientists use a method, they expect it to measure the analyte that it’s designed to measure. A method that successfully does this, without unintentionally measuring other substances too, is considered specific. Specificity is a method’s ability to measure only the target analyte, without interference from other components.
Specificity = Ability to detect only the analyte of interest
High specificity in an analytical method ensures that the obtained results reflect the product’s actual content. As a result, consumers can trust that a product is safe, its label is accurate, and what they’re consuming is reliable.
How do labs evaluate specificity?
Because specificity is so vital, scientists need techniques to assess it. Here are some strategies they use to determine if a method is specific:
- Measuring a blank sample:
The blank sample isn’t actually a sample. It’s a material that’s similar to the sample, except it doesn’t contain any analyte. Because the blank has no analyte, it shouldn’t have any quantifiable results or signal. By contrast, an analyte containing sample would be expected to show a signal. By measuring a blank, and confirming that it does not show any signal, scientists can trust that there is no interference when they measure the sample. - Measuring samples that have interfering substances:
Scientists can introduce materials (like iron or copper) that might interfere with the sample’s signal. Then, they can check the signal to see how, if at all, the material affects the analyte’s signal. This helps them understand the difference between how the sample’s signal looks when there is no interference, versus how the sample’s signal looks when there is interference. - Spiking the sample:
Scientists can add a known amount of the analyte into a sample, then measure the sample. In doing so, they will see if the method measures only that analyte.
Why Specificity Matters for Consumers
Specificity has many applications across various industries, but each application has the same common goal: to keep consumers safe. High specificity protects consumers from misleading results like data errors and false positives. This gives them confidence that the product they eat, drink, or use is safe, effective, and of high quality.
In the pharmaceutical industry, for example, a drug test must measure the active ingredient without interference from excipients or degradation products. A method’s ability to measure only the active ingredient, despite the presence of these other substances, helps guarantee that the medicine is safe and effective. In food testing, specificity ensures that laboratories correctly identify contaminants or additives that may be present in food. As a result, it prevents misleading claims about safety or nutritional content.
Specificity can also protect public health by ensuring that laboratories accurately measure environmental pollutants. For example, a scientist might use atomic absorption spectroscopy (AAS) to measure lead levels in drinking water. However, perhaps this water sample contains iron, calcium, magnesium, and zinc as well. If these metals produce signals that are close to lead’s signal, it will be hard to distinguish between (and measure) lead’s signal as opposed to the other metals’ signals. A method with high specificity ensures that iron doesn’t appear as lead in the experiment results, calcium doesn’t increase lead’s signal, and the presence of zinc doesn’t cause a false reading. In other words, high method specificity confirms that the obtained lead result is real and reliable.
Linearity
If a sample’s result depends on how much analyte it contains, then we expect that changing the amount of analyte will change the obtained result. Linearity is a method’s ability to produce results that are directly proportional to the analyte’s concentration, within a defined range.
Linearity = Proportionality between analyte concentration and experimental results
Beyond its name, linearity is more than simply “drawing a straight line.” It demonstrates a mathematical and chemical relationship, proving that the method behaves predictably and consistently across different concentrations of analyte. This proportionality is the backbone of quantitative analysis.
What linearity is truly measuring is if the instrument and chemistry obey this relationship:
Signal = m * (Analyte concentration) + b
where:
m (slope) = method’s sensitivity
b (y-intercept) = blank’s contribution or background noise
You might notice that this relationship is formatted like the slope-intercept form of a linear equation. The scientist alters the analyte’s concentration, and measures the signal (response). By measuring several samples, each of which has a different concentration of analyte, and measuring each sample’s signal, we expect a linear relationship to result.

In other words, if the analyte’s concentration doubles, then the signal should double. If the analyte’s concentration is zero, then the response should be close to the intercept. These findings indicate a proportional, linear relationship between the analyte’s concentration and the signal obtained.
What can compromise linearity?
When the relationship is not strictly proportional, what went wrong? There are a number of potential culprits:
- Instrument saturation:
Some instruments use detectors, which detect light and produce a signal based on how much light is detected. If the analyte concentration is too high, then the detector cannot absorb more light or read more signal, resulting in a flattened curve. - Chemical saturation:
At higher analyte concentrations, limits of reaction, color intensity, or ionization might cause the response to stop increasing. - Matrix effects:
Different components of the sample might produce interference. When this happens, the interference may suppress or enhance absorbance (in the case of a detector-based instrument) or alter the signal. - Contaminated standards:
The standards are sample-like materials that contain known amounts of the analyte. A method typically uses five to seven standards, each with a different concentration of analyte across the working range. To prevent interference and uphold linearity, it’s important for the standards to be prepared correctly without contamination. Wrong preparation, the presence of impurities, or degradation of these stock solutions may produce a slope that doesn’t reflect the true relationship between concentration and signal.
How do labs evaluate linearity?
Two factors help scientists quantitatively determine whether or not a method is linear:
- Coefficient of determination (R2):
In analytical chemistry, the R2 value measures the linearity of the relationship between analyte concentration and signal. A sufficiently high R2 value (close to 1.0) indicates a strong linear relationship, confirming that the calibration curve is reliable for accurate quantitative analysis. However, a high R² alone does not guarantee method accuracy; it must be considered alongside other accuracy metrics. A low R2 value, by contrast, may render the calibration invalid. - Residual plot:
A residual plot is a key tool in method validation for evaluating linearity. It reveals whether the calibration curve is truly linear, even when the R2 value appears perfect. Residuals are calculated as the actual response minus the predicted response. They should be small, randomly distributed, and free of patterns or curvature. A residual plot with seemingly random values indicates good linearity.
How to Make a Residual Plot
- Fit a linear regression (best-fit line) on the calibration data of analyte concentrations vs. measured responses.
- For each point, calculate the residual as follows: (Measured response) – (Predicted response).
- Plot residuals (on the y-axis) vs. concentration (on the x-axis).
- Draw a horizontal line at zero for reference.
Example: Linear Relationship (Good Linearity)

- Slope consistency:
Slope consistency is a way to check if the method’s response is linear and reproducible across the working range of concentrations.
- y-intercept:
In linearity discussions, the y-intercept shows the instrument’s baseline signal. Its size and consistency help assess method accuracy and reliability, particularly at low concentrations. Ideally, it should be zero, but a small non-zero value is acceptable. A small positive or negative y-intercept might result from signal interference or matrix effects. If the y-intercept is much larger than the measurement range, this might indicate systematic error, contamination of the blank, or another issue. - Quality control (QC) verification
QC verification ensures that an analytical method produces reliable and accurate results during validation and the subsequent routine analysis. To do this, scientists use QC samples with known concentrations: often low, medium, and high concentrations within the calibration range. They then compare the measured results to the nominal values to confirm that the method is performing as intended.
Why Linearity Matters for Consumers
Linearity has several purposes in analytical contexts. For instance, it ensures that the calibration curve is trustworthy and scientifically accurate. Therefore, it keeps the quantification accurate, too. With high linearity, samples’ analyte concentrations will be measured correctly, not get overestimated nor underestimated. This will be true even for samples whose analyte concentration is unknown.
As we’ve learned, even a method that is precise might still be misleading. This is where linearity becomes useful. By maintaining the accuracy and trustworthiness of the data, linearity identifies aspects of a method that might be misleading, so scientists can modify the method to improve it.
Limit of Detection (LOD)
In order to measure an analyte, the method must be able to know if the analyte is there. The limit of detection (LOD) is a parameter that represents the lowest amount of an analyte that a method can reliably detect, though they may not always quantify it accurately. By defining the LOD, laboratories ensure method sensitivity, support regulatory compliance, and enable accurate decision-making.
However, just because the method can detect the analyte, doesn’t necessarily mean that the method can quantify the analyte accurately. LOD can’t tell us in an accurate or precise way how much analyte is present. That is the job of another parameter, LOQ, which we will discuss soon.
LOD = Lowest detectable amount of analyte
The LOD is the minimum amount of analyte that the method can detect. In other words, it is the lowest concentration value that can be distinguished from background noise, though it may not be quantified accurately.
If a sample has measured analyte concentration equal to or above the LOD, then the sample’s analyte concentration is detectable and is confirmed as being present in the sample. If a sample’s analyte concentration is below LOD, then the method cannot say with certainty whether or not the analyte is present in the sample.
How do scientists know what the smallest detectable amount of analyte is? They calculate the LOD based on data that they collected from the blanks and the calibration curve:
LOD = 3.3 * (Standard deviation of blank measurements) / (Slope of calibration curve)
(3.3 is a statistical factor that represents ~99% confidence of detection)
As shown above, scientists typically define LOD as a signal that is 3.3 times the standard deviation (SD) of the blank measurements. This threshold ensures a high probability that any signal above it is real, and not random noise.
At or above the LOD, an analyte can be detected with confidence, but values below this limit are considered not detectable.
Why LOD Matters for Consumers
LOD allows laboratories to detect even the tiniest, potentially harmful substances that may be present in the food, water, or medicine. These potentially harmful substances include toxins, heavy metals, allergens, and more. The ability to detect substances like these helps confirm that products are safe and comply with strict regulatory standards. When one of these substances is detected in a sample, scientists can intervene and take action to improve the product’s quality and safety.
Lower LOD values mean that the method can detect tinier amounts of the potentially harmful substance. As a result, a low LOD lets consumers trust labels and feel confident that the products they consume do not contain dangerous trace amounts.
LOD is a parameter found widely in many different types of experiments. The examples below show how LOD may be determined in three types of experiments: AAS, high-performance liquid chromatography (HPLC), and inductively coupled plasma mass spectrometry (ICP-MS).
- AAS:
- When gold is present at a concentration of ≥0.005 mg/L in a sample, it produces a measurable signal.
- Therefore, LOD ≈ 0.005 mg/L.
- Any obtained result that is
- HPLC:
- When a pesticide is present in a sample, it produces a peak if the pesticide’s concentration is 0.1 µg/mL, but not if the concentration is 0.05 µg/mL.
- Therefore, LOD ≈ 0.1 µg/mL.
- ICP-MS:
- ICP-MS is capable of detecting signals at ng/L concentrations (parts per trillion).
- Therefore, ICP-MS methods have extremely low LODs, compared to AAS methods and HPLC methods.
Limit of Quantitation (LOQ)
Knowing that an analyte is present is one important piece of information, but another important piece is how much analyte is present. The limit of quantitation (LOQ) is the lowest analyte concentration that a method can measure with acceptable accuracy and precision, reliably distinguishing it from background noise. Unlike LOD, which only shows whether a substance is present, the LOQ indicates that the analyte can be quantified in a reliable way.
LOQ = Lowest quantifiable amount of analyte
Similarly to LOD, LOQ is calculated using the standard deviation of blank measurements and the slope of the calibration curve:
LOQ = 10 * (Standard deviation of blank measurements) / (Slope of calibration curve)
You might notice that the LOQ calculation is reminiscent of the LOD calculation, except it uses 10 where LOD uses 3.3. Therefore, LOQ is always higher than LOD. If a measured value is equal to or above LOQ, there is a very low probability that the result arises from random background noise.
At or above the LOQ, scientists can confidently report the analyte’s numerical concentration, because random error and variability stay within acceptable limits. Sometimes, a measurement may be below the LOQ, but above the LOD. These concentration values can be detected, so they are confirmed to be present in the sample, but scientists cannot quantify them reliably.
Why LOQ Matters for Consumers
LOQ ensures that scientists measure the exact amount of an ingredient, additive, or contaminant — not just detect it. By extension, it ensures that product labels stay truthful, doses remain what they should be, and safety standards are met. With a proper LOQ in place, consumers can trust that the products they use or consume are safe and consistent.
Due to its significance in consumer well-being, LOQ has far-reaching applications. Here are three examples showing how it is used across various industries:
- Pharmaceutical analysis:
- A laboratory method is designed to measure how much of a drug is in a blood sample. This method’s LOQ is 0.5 µg/mL.
- Any obtained result that is ≥0.5 µg/mL can be reported accurately and precisely.
- If the obtained result is below 0.5 µg/mL, it may be detectable if it is also above LOD, but it cannot be reliably quantified.
- Environmental testing:
- When measuring the amount of lead in drinking water via AAS, the LOQ is 5 parts per billion (ppb).
- Only water samples that have a measured lead level at or above 5 ppb can be confidently quantified, ensuring compliance with safety regulations.
- Food analysis:
- HPLC can be used to measure how much pesticide residue is present on vegetables. One such method’s LOQ is 0.01 mg/kg.
- If the pesticide level is at or above 0.01 mg/kg, it can be measured with acceptable precision.
- If the pesticide level is below 0.01 mg/kg, it may be detectable, but no quantifiable result can be reported.
- Clinical diagnostics:
- The LOQ when testing how much vitamin D is in a serum sample is 10 ng/mL.
- Only measurements at or above 10 ng/mL are reliable. Lower measurements cannot be accurately quantified.
Robustness
When scientists design experiments, they intend for the experiment to always be performed under controlled, predictable, consistent conditions. These ideal conditions aren’t always the case in reality, but the analytical method will still need to work even under less-than-ideal conditions.
The ability of an analytical method to remain unaffected by small, deliberate variations in method parameters and conditions is robustness. It measures how “sturdy” a method is when there are slight changes in temperature, pH, reagent concentrations, instrument settings, and similar conditions. Put differently, it ensures reliability during normal, routine usage.
Robustness = ability to withstand changes in experimental conditions
Robustness doesn’t mean the method is perfect under large changes, just that it tolerates minor, realistic deviations.
How do labs evaluate robustness?
Robustness is evaluated by deliberately introducing small, controlled variations in method parameters and observing the effect on results. Common parameters to vary include:
- Instrumental variables: e.g., wavelength, column temperature, flow rate (for chromatography)
- Chemical variables: e.g., pH of buffer, reagent concentration, mobile phase composition
- Operational variables: e.g., extraction time, mixing time, injection volume
Note that, in this context, “method parameters” doesn’t refer to the method validation parameters we’ve discussed (accuracy, precision, specificity, etc.). Instead, it refers to properties of the method itself that determine how the method is performed.
How to Set up a Robustness Experiment
- Based on the method’s sensitivity, select critical parameters of interest.
- Alter each parameter slightly from its typical value (e.g., ±2°C in temperature, ±0.1 pH unit, ±5% reagent concentration).
- Measure the standards and samples as they would normally be measured.
- Use the results to calculate spike recovery, precision, and accuracy.
- Assess whether or not the slight alterations in parameter caused a significant deviation from the normal results.
If there was not a significant deviation, then the method is robust. In other words, it is considered resilient and reliable under routine laboratory conditions.
Why Robustness Matters for Consumers
It’s normal for laboratory conditions to change slightly, even on a daily basis. Having a robust method ensures that, when these conditions do change, the integrity of the experiment’s results will be just as accurate and trustworthy. Thus, robustness is key to all analytical lab testing.
In the pharmaceutical industry, robustness ensures that drug content is accurately measured, even if small changes occur in temperature, pH, or instrument settings. This guarantees safety and efficacy. In the food industry, it assures scientists that nutrient or contaminant levels are reliably reported, despite minor variations in testing conditions. Finally, in environmental testing, it’s thanks to robustness that pollution or toxin measurements remain accurate, enabling scientists to safeguard environmental and public health.
Why does method validation matter for consumer safety?
